This introduction presents an overview of the key concepts discussed in the subsequent chapters of this book. The book explains how deoxyribonucleic acid (DNA) sequence information is obtained and analyzed, and some of the major reasons for doing so. It describes the sequencing methods in common usage, with particular emphasis on the relative merits and pitfalls of each approach. The book also describes the major applications of these methods. It discusses the computer-based analysis of sequence data. DNA sequencing is the determination of the base sequence of all or part of a DNA molecule. The base sequence is just a part of the complete description of the structure of a DNA molecule. In addition to restriction endonucleases, many other nucleic acidbinding proteins show at least some sequence specificity. These include the transcription and translation machinery responsible for interpreting the protein-coding information encoded in the DNA.
